基于ABCG3l1探讨电针治疗大鼠脑卒中后认知功能障碍的作用机制Exploration of the mechanism of electroacupuncture in treating post-stroke cognitive impairment in rats based on ABCG3l1

摘要(Abstract):
目的:观察电针百会、神庭穴对脑卒中后认知障碍(PSCI)模型大鼠的神经功能缺损情况、学习记忆功能和ABCG3l1的mRNA相对表达的影响,探讨其改善PSCI的作用机制。方法:将27只雄性SD大鼠按随机数字表法分为假手术组、模型组和电针组,每组9只。采用Zea Longa改良线栓法制备大脑中动脉栓塞(MCAO)模型,造模后第1天对电针组进行电针百会、神庭穴干预,20 min/次,1次/d,持续7 d。假手术组和模型组同等条件抓取,不进行电针干预。采用ZeaLonga 5分法对大鼠进行神经功能缺损评估,Morris水迷宫实验评估大鼠空间学习和记忆能力,实时定量荧光PCR(RTqPCR)检测海马组织中ABCG3l1的mRNA相对表达量,生物信息学方法分析ABCG3与ABCG3l1的同源性。结果:生物信息学分析得知ABCG3与ABCG3l1具有高度同源性;Zea-Longa 5分法中,造模后第1天、第7天,模型组和电针组神经功能评分均较假手术组升高,差异有统计学意义(P<0.01);与模型组造模第7天和电针组第1天比较,电针组第7天神经功能评分均明显下降,差异有统计学意义(P<0.05)。在Morris水迷宫定位航行实验中,重复测量方差分析结果显示,组别主效应、干预时间主效应、组别与干预时间的交互效应均显著。Morris水迷宫空间探索实验检测结果表明,与假手术组比较,模型组大鼠的平台穿越次数明显减少,差异有统计学意义(P<0.05);第4、5、6天测试数据显示,与假手术组比较,模型组大鼠平台探索总时长增加,差异有统计学意义(P<0.01);与模型组比较,电针组大鼠平台探索总时长缩短,差异有统计学意义(P<0.01)。在RT-qPCR实验中,与假手术组比较,模型组ABG3l1 mRNA表达量明显增高,差异有统计学意义(P<0.05);与模型组比较,电针组ABCG3l1 mRNA表达量明显降低,差异有统计学意义(P<0.05)。结论:电针百会、神庭可以改善PSCI模型大鼠的神经功能评分,提高大鼠空间学习和记忆能力,降低缺血侧海马组织中ABCG3l1的mRNA相对表达,改善PSCI大鼠的认知障碍,其机制与纠正海马组织中ABCG3l1的mRNA表达有关。
