疏风解毒胶囊对急性咽炎模型大鼠TLRs/MyD88信号通路的影响Effects of Shufeng Jiedu Capsule on the TLRs/MyD88 signaling pathway in model rats with acute pharyngitis

摘要(Abstract):
目的:探究疏风解毒胶囊对急性咽炎(AP)模型大鼠的抗炎效果及其对TLRs/MyD88信号传导途径的作用。方法:选取50只健康清洁级SD成年大鼠,按随机数字表法分为7个组,包括空白对照组7只、造模组7只、安慰剂组6只、阿莫西林组6只,以及低、中、高3个不同剂量疏风解毒胶囊治疗组各6只,造模过程中耗损6只。使用喉头喷雾装置对大鼠咽部进行15%浓度氨水喷洒处理,构建AP动物模型,造模成功后给药;空白对照组以等量生理盐水处理。自第4天起灌胃干预:疏风解毒胶囊组按临床等效剂量换算为2.2 g/(kg·d),1.1 g/(kg·d),0.55 g/(kg·d)灌胃,阿莫西林组给予20 mg/(kg·d)灌胃,安慰剂组予等量蒸馏水灌胃。2次/d,持续5 d。病理切片进行苏木精-伊红染色法(HE)染色,观察大鼠咽部黏膜的形态学改变。采用酶联免疫吸附测定(ELISA)技术测定各组大鼠血清中IL-6、IL-10及TNF-α的含量表达。使用聚合酶链逆转录(RT-qPCR)技术,对咽部组织中的TLR2、TLR4以及MyD88的mRNA表达水平进行定量检测。同时,利用ELISA进一步分析咽部组织中TLR2、TLR4和MyD88蛋白的表达比例。结果:疏风解毒胶囊组可改善氨水诱导的AP大鼠咽部组织病理学改变,减少炎症细胞浸润。与安慰剂组比较,阿莫西林组与疏风解毒胶囊各剂量组IL-6、TNF-α水平、TLR2、TLR4、MyD88蛋白表达及其mRNA表达水平均降低,IL-10水平均升高,差异有统计学意义(P<0.05)。与阿莫西林组比较,疏风解毒胶囊中、低剂量组大鼠血清IL-6、TNF-α水平较高,IL-10水平较低,TLR2、TLR4、MyD88蛋白的表达水平及mRNA表达水平均升高,差异有统计学意义(P<0.05)。与疏风解毒胶囊低剂量组比较,中剂量组及高剂量组IL-6、TNF-α水平及TLR2、TLR4、MyD88蛋白的表达水平及mRNA表达水平均较低,IL-10水平升高,高剂量组IL-6、TNF-αIL-10水平变化更明显,TLR2、TLR4及MyD88蛋白的表达水平及mRNA表达水平均低于中剂量组,差异有统计学意义(P<0.05)。结论:疏风解毒胶囊可能通过调控TLRs/MyD88通路及炎症因子表达发挥抗炎作用。
